Hello, I am new to KiCad and have version 5.0
I have a rather thick substrate (0.250") and want to define a volume that will be drilled to a set depth with a diameter that is relatively large (0.50"). I do not want a through hole.
Is this possible in KiCad?

Not directly. You have to ask your board manufacturer if they can do it and how they want the information. Most probably it would be just another gerber layer which shows the location of the drill.
